Abstract 15120: Attenuated Renal Injury After the Post-Resuscitation Administration of Doxycycline in Surviving Newborn Piglets With Severe Hypoxia-Reoxygenation

Background: Asphyxiated neonates often have multi-organ failure with myocardial dysfunction and renal insufficiency. Previously we demonstrated that doxycycline (DOX) improved cardio-renal function in an acute swine model of neonatal hypoxia-reoxygenation (HR). To examine whether the cardio-renal protective effects of intravenous (iv) DOX persisted in a surviving model of neonatal HR. Design: Piglets (1-4 days-old, weighing 1.7-2.4 kg) were instrumented with catheters placed in common carotid artery and external jugular vein for hemodynamic monitoring and iv administration of medications, respectively. Piglets were subjected to 1 hour of hypoxia followed by reoxygenation with 21-25% oxygen and observed for 4 days. An iv bolus of DOX (30 mg/kg) or normal saline (control) was given at 5 minutes of reoxygenation in a blinded, block-randomized fashion (n=8/group). Sham-operated piglets (n=6) received no HR.
