Preliminary Studies on Regeneration Capacity of Plum (Prunus Domestica L.) Somatic Tissues in Vitro

ABSTRACTIn vitro regeneration of two plum genotypes was investigated starting from different explants—leaf, stem and root segments. Plants were regenerated successfully from leaf explants. We established as optimal regeneration medium MS-30 basal medium with addition of 0.5 mM IBA and 7.5 mM TDZ. The regenerating shoots developed directly from the explants without intermediate callus phase. Shoots were produced in 46.25 per cent for cv. Stanley and 31.43 per cent for cv. Kjustendilska sliva of the explants. This is the first report on successful regeneration of plums from leaves and it is a new perspective for application of genetic transformation.
