Detection of Serum Biomarkers for Different Symptom Type of Knee Osteoarthritis Using Surface Enhanced Laser Desorption/Ionization Protein Chip System

Objective:New technologies for the study of knee osteoarthritis(OA) proteomic are urgendy needed.The aim of this study was to screen serum biomarkers in patients with knee OA by using surface-enhanced laser desorption and ionization time-of-flight mass spectrometry(SELDI-TOF-MS)technique.Methods:Proteomic spectra were generated by mass spectrometry in 105 cases,including 30 cases of deficiency of liver and kidney,35 cases of stagnation of vital energy and blood stasis.and 40 cases of healthy control.The discriminatory profiling between knee OA and normal samples was analyzed by Biomarker Wizard software and Biomarker Pattern softwar.Results:eighteen differentially expressed proteins in serum were screened by analysis of protemic spectra of knee OA of deficiency of liver and kidney and healthy control(P0.01).13 proteins in knee OA of deficiency of liver and kidney were increased and 5 proteins in healthy controls were increased.twenty differentially expressed proteins in serum were screened by analysis of protemic spectra of knee OA of stagnation of vital energy and blood stasis and healthy control(P0.01).6 proteins in knee OA of stagnation of vital energy and blood stasis were increased and 14 proteins in healthy controls were increased.5 differentially expressed proteins in serum were screened by analysis of protemic spectra of knee OA of deficiency of liver and kidney and stagnation of vital energy and blood stasis(P0.01).Concluion:SELDI-TOF-MS offers a unique platform for the proteomic detection of different symptom type of knee OA.It also offers a noninvasive method to further study the proteomic changes in the development and progression of knee OA.
