Influence of high glucose on expression of CD44 in human retinal pigment epithelial(hRPE) cells in vitro

Objective: To investigate the influence of high glucose on expression of CD44 in human retinal pigment epithelial(hRPE) cells in vitro.Methods: Proliferation of hRPE cells cultured with 5.6mmol/L and 30.0 mmol/L of glucose for 24,48and 72h were measured by MTT colorimetry.Expression level of CD44 in hRPE cells with 5.6 mmol/L and 30.0 mmol/L of glucose were measured by flow cytometer.Result: The proliferative inhibition on the surface of hRPE cells cultured with 30.0 mmol/L of glucose for 24,48 and 72h were 11.4%、6.20%、5.36%,respectively comparing with that cultured with 5.6 mmol/L of glucose.The expression of CD44 in RPE cells cultured with 30.0 mmol/L glucose was 40.39%,while only a little quantity of CD44 expression was detected in REP cells cultured in 5.6 mmol/L showing significant difference between the two groups(P0.05).Conclusions: High glucose can inhibit the proliferation of RPE cell in vitro,while increase the expression of CD44.
