The invention relates to a one-dimension silver chain nano-structure, a self-assembly preparation method and an SERS application. The self-assembly preparation method includes the following steps that (1) silver nano-particles are scattered in an alcohol-water mixed solution containing inorganic salt, and then a solution A is obtained and subjected to standing; and (2) a certain amount of alkaline solution or an alkaline-containing alcohol solution is added into the solution A, stirring and centrifugal separation are conducted, and then the one-dimension silver chain nano-structure is obtained. The one-dimension silver chain nano-structure has the good SERS effect. The one-dimension nano-structure with the silver nano-particles of different sizes and different lengths is obtained by controlling the static interaction force among the silver nano-particles in a self-assembly mode. According to the method, any templates and surface active agents are not needed; the assembly process is simple; the sizes of the silver nano-particles are easy to control; the product is likely to be separated; the SERS effect is remarkable; a preparation system is environmentally friendly; no toxic or harmful solution is added; and the prepared product is stable in performance, low in preparation cost, wide in application range, good in repeatability and easy to prepare in large scales.
