Effect of Different Hypoxia Concentration Exposure on γδT Cells within Chronic Fatigue Syndrome Mice

Objective: It is to study the effect of different hypoxia concentration exposure on γδT cells expression in intestine in mice with chronic fatigue syndrome( CFS). Methods: 72 ICR male mice were randomly divided into 6 groups( 12 per group),the model control group( C 0),the chronic fatigue syndrome model group( CFS 0),the hypoxia intervention control group( C),the hypoxia intervention control group with chronic fatigue syndrome( CFS), the chronic fatigue syndrome with 15% O 2 concentration hypoxia exposure intervention group( 15% H) and the chronic fatigue syndrome with 18% O 2 concentration hypoxia exposure intervention group( 18% H). Among them,the CFS 0 group,CFS group,15% H group and 18% H group were induced by restraint and forced swimming,and C 0 group and C group were fed in the same condition. After the establishment of CFS model,C 0 group and CFS 0 group were used to test the effect of CFS model. C group continued to feed for 7 days,and CFS group recovered for 7 days; 15% H group and 18% H group were respectively exposed to 15% O 2 concentration hypoxia and 18% O 2 concentration hypoxia for 7 days( one hour per day). The number of γδT cells in intestine was examined by the flow cytometer. Results: 1) There were no significant difference in the body weight between C 0 group and CFS 0 group during the first week of the establishment of mice model,but body weight in CFS 0 group was significantly lower than that in C 0 group in the following 2,3,4,5 weeks. 2) Morris water maze( MWM) results showed that the incubation period of CFS 0 group was shorter compared with the C 0 group. In addition,the platform quadrant time in CFS 0 group was longer compared with C 0 group. 3) The percentage of γδT cells in CD3+iIEL cell( intestinal intraepithelial lymphocytes) in CFS group and 18% H group decreased significantly,compared with C group. however, the percentage of T cells in CD3+iIEL cell in 15% H group was significantly higher than that of CFS group and 18% H group,and had no difference with C. Conclusions: Based on the change in the general body weight and behavioral indicators,the model of chronic fatigue syndrome has been successfully established by restraint and forced swimming. Moreover,the exposure to 15% O 2 hypoxia for 7 days( 1 hour / day) can significantly improve the expression of γδT cells in intestine in the chronic fatigue syndrome.
