Functional expression of the mutants of the chloroplast tRNALys gene from the liverwort, Marchantia polymorpha, in Escherichia coli

The anticodon of the tRNALys gene (trnK) in the liverwort, Marchantia polymorpha, was anificially converted to an amber anticodon. This mutant tRNALys(CTA) gene carrying either the intron or the C27-C43 mismatch at the anticodon-stem is not functional in Escherichia coli, but without both of them, it does work as a tRNALys amber suppressor.
