EFFECTS OF GDNF AND HSV-GDNF ON Bcl-2 EXPRESSION OF THE DEVELOPMENT OF MUSCLE SATELLITE CELLS OF NEWBORN RAT IN VITRO

In order to investigate the effect of GDNF and HSV-GDNF on newborn rat satellite cells, myoblasts and myotubes of skeletal muscle ,we observed the expression of Bcl-2 on these cells in vitro. We established three groups (control, GDNF, HSV-GDNF group) after the third passage. The skeletal muscle cells were cultured for 2, 4, 8, 16, 24, 48, 72 h individually. Then these cells were stained with Bcl-2 monoclone immunohistochemical staining technique. The positive Bcl-2 immunoreactivity was present in the satellite cells, myoblast cells of skeletal muscle, but not in myotubes. It showed strongly in groups pretreated with GDNF and HSV-GDNF; and the number of the living cells is also more than that in the control group. The present results suggest that GDNF and HSV-GDNF might inhibit apoptosis, promote clonal expansion and differentiation on the cells of skeletal muscle mentioned above in virto.
