Interaction between Carbonic Anhydrase and Its Inhibitors Studied by Means of A pCO2 Electrode

Abstract A simple and reliable method for determining the activity of carbonic anhydrase, in equilibrium conditions, is presented. The method is based on the linear variation of a pCO2 probe electrode potential which follows the CO2 evolution, a first order reaction. Different experimental conditions (geometry of the system, temperature, pH) are considered. The method allowsalso the study of inhibition or activation displayed on carbonic anhydrase by different compounds in conditions of temperature, concentration, pH etc. fixed and controlled. In particular the possibility of performing the measurements in condition of constant pH, seems to be one of the main advantages of such method.
