The invention discloses a method for detecting giant knot weed metabolic substance group in rat plasma based on combination of molecular imprinting nanometer magnetic bead dispersive solid-phase extraction and LC/Q-TOF-MS technology. The method comprises the steps that imprinting magnetic beads are prepared, the prepared imprinting magnetic beads are added into processed rat plasma for adsorption and analysis, and an analysis solution is collected; the analysis solution is blown to be dry through nitrogen, a flow phase and reference substance genistein are used for redissolving, the LC/Q-TOF-MS is utilized for carrying out semi-quantitative analysis on the analysis solution, and the metabolic substance group of the traditional Chinese medicine giant knot weed is identified. By the adoption of the method, based on the function characteristic that biological sample substrate is complex, the relevance of original ingredients, the metabolic substance group and activity of the traditional Chinese medicine giant knot weed is disclosed by enriching the metabolic substance group, combining activity screening and tracing the traditional Chinese medicine original ingredients, the number of target metabolic products obtained through identification is large, and the enrichment effect is good.
