Study on New Preparation Technology of Maize Resistant Starch by Autoclaving-Enzyme Method

Maize starch was used as raw material,the method which combined autoclaving-cooling(120℃,30min then 4℃,24h) circle with pullulanase hydrolysis was used to prepare resistant starch(RS3).Treatment times of autoclaving-cooling circle was studied.The optimal hydrolysis condition of pullulanase,including pullulanase concentration,hydrolysis temperature and time was determined by orthogonal test.The results showed that the method which added pullulanase after starch gelatinization and hydrolyzed at the condition of selected pullulanase concentration(4.0U·g-1),hydrolysis temperature(55℃),pH value(4.5),and hydrolysis time(8h),then cooled the starch for retrogradation and then treated it by twice autoclaving-cooling circle,may significantly increase the yield of resistant starch from maize starch.
