Influence of Fat Acid on the Expression of Adiponectin Receptor 1 and 2 of HepG2 Cell

Objective To investigate the function of adiponectin receptor in the process of insulin resistance induced by high free fat acid.Methods HepG2 hepatic cell was cultivated in DMEM with 10% FCS for 48 hours,then treated with 100μmol/L,200μmol/L and 400 μmol/L palmitic acid(PA) separately,incubated for 24 hours continuously.Half-quantitative of the whole RNA and RT-PCR were extracted to analyze the expression of adiponectin receptor,and compare with the control group without PA.Results Compared with the control group,R_2 expression of HepG2 decreased in the group with PA.When the concentrate of PA was over 200 μmol/L,R_2 expression decreased significantly difference compared with control and PA 100 μmol/L group (P0.01).In all the groups,R_1 expression also represented the decline tendency,but there is no significant difference(P0.05).Conclusion The decrease of R_2 expression in hepatic cell may participate in IR induced by FFA.
