Design and Synthesis of Some Phenoxy Acids as Potential Non-Covalent Antisickling Agents

Two series of phenoxy acids, 18 compounds (Fig. 1), were designed and synthesized to be evaluated as antisickling agents. Oxygen equilibrium study of the target compounds was performed using Oxygen Equilibrium Curve (OEC). Some of the tested compounds causedleft-shift the allosteric equilibrium to produce a high-O2-affinity Hb, others showed right-shift the curve indicating low-O2-affinity Hb, while the rest did not affect the Oxygen Equilibrium Curve. RBC morphological antisickling study of representative compounds 7b (left shifted) and 17d (right shifted) were also performed.The obtained results indicated more RBC sickling effect, as expected for 17d, while no effect was observed for 7b.
