Abstract 17541: SDPR/Cavin-2 Modulates Apoptosis via PTEN/Akt Signaling Pathway and Deteriorates Cardiac Function in Pressure Overload-Induced Heart Failure

2017
Introduction: Cavins and caveolins are the principal membrane components of caveolae. Some of cavin family proteins affect cardiac function. Although the structural role of SDPR/Cavin-2 has been well elucidated, the functional role of SDPR/Cavin-2 remains unknown. Objective: We investigated the functional role of SDPR/Cavin-2 in heart failure induced by pressure overload. Methods and Results: To examine the role of SDPR/Cavin-2 in cardiac function, transverse aortic constriction (TAC) was performed on WT mice and SDPR/Cavin-2 deficient mouse (SDPR/Cavin-2 KO) mice. Four week after TAC procedure, echocardiography showed that left ventricular fractional shortening (LVFS) and left ventricular ejection fraction(LVEF) was significantly preserved in SDPR/Cavin-2-/- mice compared with WT mice. (%FS; 27.4 ± 2.0% in SDPR/Cavin-2 KO vs. 22.4 ± 0.95% in WT, p<0.01, %LVEF; 53.8 ± 3.1% in SDPR/Cavin-2 KO vs. 45.7 ± 1.8% in WT, p<0.01). Cardiomyocytes with adenovirus-mediated SDPR/Cavin-2-knockdown were resistant to ap...
