Production of transformed cell of vertebrate animal and its cell culture

1995
PROBLEM TO BE SOLVED: To obtain the subject transformed cell for polypeptide replication by transducing a manifestation vector containing a vector fragment capable of replicating in a host cell, a promoter having compatibility with the host cell and a polypeptide DNA into a cell of a vertebrate animal. SOLUTION: A DAN transfer vector fragment capable of replicating in a host cell culture, a DNA fragment containing a promoter having compatibility with a host cell culture and a DNA fragment which codes a polypetide and is linked to the promoter but is different from the species of the promoter are combined with a transcription termination site placed at a proper position, a poly A addition sequence and translation initiation signal and stop signal to give a manifestation signal, which is under control of the promoter or is formed in such a way that a translation initiation codon exists at an essential position in which the translation initiation codon is occupied by a translation initiation codon of a protein under control of the promoter. The manifestation vector is transduced into a cell culture of vertebrate animal and transformed to give the objective transformed cell culture of vertebrate animal for polypeptide replication.
