Study on Rabbit Plasma Protein Binding Rate of Nitidine Chloride

OBJECTIVE: To determine the rabbit plasma protein binding rate of nitidine chloride. METHODS: The equilibrium dialysis combined with HPLC was used to determine the rabbit plasma protein binding rate of nitidine chloride. RESULTS: Nitidine chloride and the internal standard (chloromycein) were separated completely without interference. The linear range was 0.031 88~2.040 00 μg·mL-1. Its recovery rates were more than 95% at the concentrations of 0.125 μg·mL-1, 0.25 μg·mL-1, 0.5 μg·mL-1. The RSD of intra-day and inter-day both were less than 5%. The plasma protein binding rates of nitidine chloride with concentrations of 0.125 μg·mL-1,0.25 μg·mL-1 and 0.5 μg·mL-1 were 73.21%,72.83%,70.30%, respectively. CONCLUSION: Nitidine chloride shows moderate rabbit plasma protein binding rate.
