5-Lipoxygenase and Phospholipase A2 Inhibitory Potentials of Alcoholic Extract of Cyperus rotundus: In vitro and In silico Study

The literature revealed that not much reports are known for the anti-inflammatory activity of any Cyperus rotundus plant part (or extract) with respect to the molecular targets (inflammatory mediators) like cyclooxygenase-1/2 (COX-1/2), 5-lipoxygenase (5-LOX), and phospholipase A 2 (pLA 2 ). The present investigation involved exploration of in vitro inhibitory potential of alcoholic extract of C. rotundus rhizome against two important inflammatory mediators; 5-LOX and (porcine) pLA 2 . In addition to it, the role of phytoconstituents, and the key chemical constituents present in the extract for mediating the anti-inflammatory activity was explored by in silico studies, utilizing the Schrodinger Maestro 9.1 software. The present study represented that the alcoholic extract of C. rotundus exhibited low 5-LOX and ppPLA 2 inhibitory potential with 26% and 29.5% inhibition, respectively. The in silico studies revealed that the phytoconstituents myrtenol (by strongly binding with histidine residues and Fe 2+ ) and germacrene B (by strongly interacting with isoleucine and phenylalanine residues) demonstrated the highest G score due to the formation of hydrogen bonding. The research opened the perspective of proving the ethnopharmacological utility of C. rotundus in traditional medicine for treating inflammatory conditions. The in silico study of probable phytoconstituents and their mechanism will help in isolating, optimizing, and further use for developing better anti-inflammatory agents in coming future.
