MANGIFERIN PROTECTS HEART FROM ISOPROTERENOL-INDUCED MYOCARDIAL NECROSIS IN RATS

Objectives: To investigate the cardioprotective potential of Mangiferin, isolated from leaves of Mangifera indica (Anacardiaceae), in isoproterenol (ISO) induced myocardial necrosis in rats. Methodology: Male albino Wistar rats were divided into six groups: 1-Normal; 2-ISO-control; (3-5)-Mangiferin (10, 20 & 40 mg/kg; i.p.); 6-Mangiferin per se. Mangiferin or vehicle was administered to rats for 15 days.  On 13th and 14th day, ISO (85mg/kg; s.c.) was administered to rats at an interval of 24 h. Results: Administration of ISO resulted in significant decrease in arterial blood pressures (SAP, MAP and DAP), heart rate (HR) and maximum positive and negative rate of left ventricular pressure (±LVdP/dtmax) with concomitant increase in left ventricular end-diastolic pressure (LVEDP) (p<0.001). Furthermore, there was marked rise in malondialdehyde, a lipid peroxidation marker; increase in cardiac injury enzyme activities in serum and depletion of antioxidant status. Histopathological studies revealed necrosis, edema and infiltration of inflammatory cells in the heart tissue. In contrast, pretreatment with mangiferin (40 mg/kg; i.p.) significantly prevented the cardiac necroenzyme release, decreased necrosis and inflammation, improved cardiac function and restored myocardial oxidant-antioxidant status. Conclusion: The present study suggests that mangiferin exerts cardioprotective effect in ISO-induced myocardial injury via upregulation of antioxidant defense systems.
