Impact of polymorphisms of the DNA repair gene XRCC1 and their role in the risk of prostate cancer.

Objective: Weconductedacase-controlstudytoexaminetheroleofXRCC1�codons�194�(Arg>Trp),�280� (Arg>His)�and�399�(Arg>Gln)�polymorphismsintheriskofprostatecancer. Methods:�Thisstudyincluded�572�consecutiveprimaryprostatecancerpatientsand�572�controlsbetween� January�2011�andJanuary�2014.�Thepolymerasechainreaction-restrictionfragmentlengthpolymorphism� (PCR-RFLP)� wasperformedtodetectXRCC1� codons� 194� (Arg>Trp),� 280� (Arg>His)� and� 399� (Arg>Gln)� polymorphisms. Results: Comparedwiththecontrolsubjects,�theprostatecancercaseshadahabitofcigarettesmoking� (χ 2 =18.13,� P Trp)�polymorphismandclinicalstageandGleason� scoreofprostatecancer�(P>0.05). Conclusion:�OurresultsshowanincreasedriskforprostatecancerinindividualswithXRCC1�194�(Arg>Trp)� polymorphism,�andasignificantinteractionbetweenXRCC1�194�(Arg>Trp)�polymorphismandtobacco� smoking,�alcoholdrinkingandfamilyhistoryofcancer.
