Detection and identification of Vibrio parahaemolyticus by matrix assisted laser desorption/ionization time of flight mass spectrometry.

Objective To establish a rapid method for the detection and identification of Vibrio parahaemolyticus by matrix assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS).Methods In order to verify the stability and repeatability,the reference strain V.parahaemolyticus cultured in different medium or different time and the isolates which had been cryopreserved for a long time were detected by MALDI-TOF-MS.The strains from different sources had also been tested to confirm the accuracy of this method.Results Different medium such as PW,TCBS and Vibrio colored medium and different culturing time interval of 18,42 or 72 h had little impact for the mass spectrometry of the bacteria.There were no changes for the identification scores of the bacteria which had been preserved for two years using MALDI-TOF-MS.The strains from different countries and regions get the same accurate results.Conclusion MALDI-TOF-MS is a fast and accurate method for the identification of V.parahaemolyticus by MALDI-TOF-MS,and it can be used for routine detection because of its good stability and repeatability.
