Study on the dosage of AccQ.Flour reagent in the test of determination of amino acids by pre-column derivatization

Objective: To find out the best dosage of derivative reagent which used for the different concentrations of standard solutions of amino acids.Methods: Taked the highest concentration of amino acid sample which can derived by 20 μl derivative reagent,and reducted derivative reagent dosage followed to find out the the best dosage of derivative reagent which used for the different concentrations of standard solutions of amino acids according to mathematical statistical analysis.Results: The peak area of the 0.75 μmol/ml amino acid standard solution derived by 17 μl derivative reagent was not significant different with the peak area derived by 20 μl derivative reagent.The peak area of the 0.5 μmol/ml amino acid standard solution derived by 15 μl derivative reagent was not significant different with the peak area derived by 20 μl derivative reagent.Conclusion: The best dosage of derivative reagent which used for the standard solutions of amino acids which concentration not over 0.5 μmol/ml is 15 μl;the best dosage of derivative reagent which used for the standard solutions of amino acids which concentration not over 0.75 μmol/ml is 17 μl.
