The invention discloses a ganoderic acid high yield engineering strain kmust-SE and a construction method thereof. The engineering strain is preserved in China General Microbiological Culture Collection Center, CGMCC, and the preservation number is CGMCC No. 11401. The provided engineering strain (kmust-SE) is prepared through overexpression of SE gene that encodes squalene monooxygenase by lucid ganoderma strong promoter P-gpd. The shake-flask fermentation experiment results show that the contents of monomer ganoderic acid GA-Me, GA-T, GA-Mk, and GA-S, which are produced by SE converted sub-strain, are 1.2, 2.1, 2.5, and 1.7 times higher than those of WT strain. The high yield strain can be used as the engineering strain for producing ganoderic acid, and the application prospect is wide.
