Effect of Light on the Callus Induction of Lavandula Latifolia

Objective:OBJECT Probe the effect of light and darkness condition on the callus induction of Lavandula latifolia Vill.METHOD Leaves and stems of Lavandula latifolia taken as explants,the callus was inducted and cultured in two different medium which are(1)MS+2,4-D1.0 mg/L+NAA0.3 mg/L+KT0.2 mg/Land(2)MS+2,4-D0.1 mg/L+6-BA0.5 mg/L.RESULT In the light,the callus induction rates of leaves and stems are both 100% in the(2) medium with the light green callus being fine and close.And the callus induction rates are both 88.57% in the(1) medium with the dark gray callus from leaves being loose and the light green callus from stems being fine and close.In the darkness,the callus appeared grayish white,loose and soaked.The callus induction rate of leaves and stems are 100% and 88.57% respectively in the medium(2) and both only 80% in the medium(1).CONCLUSION Light is better for the callus induction of Lavandula latifolia.The optimum medium for Lavandula latifolia.L is(2) MS+2,4-D0.1 mg/L+6-BA0.5 mg/L.And the differences of the callus are not significant whether leaves or stems are selected as explants.
