Tumor suppressor gene screening on the long arm of chromosome 1 in Chinese patients with sporadic colorectal cancer

Objective To screen out the potential tumor suppressor gene (TSG) on D1S413-D1S2622 in Chinese patients with sporadic colorectal cancer.Methods On the basis of our previous study,we chose 25 genes located in the D1S413-D1S2622 region and presented a microarray-based high throughput screening approach in 19 sporadic colorectal cancer cases to identify candidate colorectal cancerrelated tumor suppressor genes.The relationship between expression levels of candidate genes and the clinicopathological data was analyzed.Results According to the microarray-based high throughput screening,we found 4 significantly down-expressed genes,including CSRP1,LMOD1,PPP1R12B and CFHL3,which were found to be down-regulated in 15,16,16,and 16out of the 19 cases,respectively.Each of them was down-regulated over 2-fold in 11,11,14 and 10 samples,respectively.There was no significant association between expression levels of CSRP1,LMOD1,PPP1R12B,CFHL3 and the clinicopathological data.By database searching,CSRP1 was hypothesized to be a colorectal cancer related tumor suppressor gene.Conclusion Through microarray-based high-throughput screening and subsequent database searching,we present the evidence that CSRP1 might be involved in the progression of colorectal cancer.
