Costimulation molecule B7-H7 expression effects biological function human glioma cell lines

Objective To study the expression of B7-H7, a co-stimulation molecule, in human glioma glioma cell line. Methods Down-regulation of B7-H7 was performed by using RNA interference (RNAi) method in human glioma cell line U87 and U251. Results By using the RNA interference technology, we successfully decreased B7-H7 expression in human glioma cell lines. Cell counting kit-8 (CCK-8) cell proliferation experiment showed that in 48 h and 72 h, the proliferation rate of U87-LV-shB7-H7 cells was significantly lower than that of U87-LV-NC (48 h: t=2.826, P<0.05; 72 h: t=3.707, P<0.05), and in 24 h and 72 h, the proliferation rate of U251-LV-shB7-H7 cells was significantly lower than that of U251-LV-NC (24 h: t=3.479, P<0.05; 72 h: t=3.311, P<0.05). In wound healing assay, the cell-free areas of the LV-shB7-H7 groups were significantly wider than that of LV-NC groups, after 24 h (U87: t=5.966, P<0.01; U251: t=4.034, P<0.05). In Transwell invasion assay glioma cells from LV-shB7-H7 groups showed significantly reduced number of cells invading through the matrigel, as compared to cells from LV-NC groups (U87: t=10.660, P<0.01; U251: t=5.442, P<0.01). Conclusion Expression of B7-H7 plays an important role in the regulation of the biological behavior of glioma cell lines. Key words: B7-H7; Glioma; U87; U251; RNA interference
