Effects of pp32 on prostate cancer cells

Objective To explore the effect of pp32 on prostate cancer cells. Methods The pcDNA3. 1 and pcDNA3. 1-pp32 plasmids previously stored in our laboratory were transferred into Pc3M cells in order to construct stable Pc3M-pp32 cell lines. Morphological changes of Pc3M cells were observed by Giemsa staining experiments,and the cells ability of migration was measured through Transwell's chamber assays. The cells ability of adhesion was assessed by adhesion experiments,and MTT assays were used to determine the cell growth curve. Finally,the expression of N-cadherin was investigated by Western blotting. Results We successfully obtained Pc3M cell lines with pp32 over-expression(Pc3Mpp32) ,and pp32 significantly induced morphological changes of Pc3M cells. Pp32 could also enhance the migration and adhesion of human prostate cancer cells Pc3M. Pp32 could obviously increase Pc3M cell growth and effectively inhibit the expression of N-cadherin in Pc3M cells. Conclusion Over-expression of pp32 leads to morphological changes of Pc3M cells,and promotes the migration,adhesion,and growth of these cells.
