A Screen for Molecular Markers of Cisplatin Resistance in Glioma Cells

BACKGROUNDOBJECTIVE:Cisplatin is one of the most common used chemotherapy agents in neuro-oncology.Drug resistance could be an important obstacle resulting failing of chemotherapy.Genetic changes may affect the chemosensitivity of cancer cells.In this study,we try to screen molecular markers of cisplatin resistance in glioma cells.METHODS:Array comparative genomic hybridization(aCGH) aCGH have been used to screen molecular markers of cisplatin resistance using glioma cell line U251 and cisplatin induced resistant cell line U251/CP2(IC50 to cisplatin were 1.2 μg/ml and 76.5 μg/ml,respectively) .RT-PCR and real time PCR were used to verified the results of aCGH.RESULTS:The U251 cells have showed lost at least one copy of each complement factor H-related protein 1(CFHR1) and complement factor H-related protein 3(CFHR3) genes,and both CFHR1 and CFHR3 are homozygously deleted in U251/CP2.While U251/CP2 cells have gained 2 ~3 copies of IL-7 as compared with parent line U251.CONCLUSION:CFHR1,CFHR3 as well as IL-7 may relate with cisplatin resistance in human gliomas.
