Effect of carbon sources on activity of α-amylase by B.subtilis

The addition of glucose,sucrose and starch to the chemically defined medium,which was devised for use in the growth,pH and α-Amylase induction studies.As a result,the inoculated flask was incubated on a rotary shaker,the activity ofα-Amylase reached the highest point on 54h.When starch was used as a carbon source,the maximum activity ofα-amylase reached(114.5±2.3)U/mL,the cell density was 2.342±0.023.Then when glucose,sucrose were respectively used as a carbon source,the activity ofα-amylase were(75.6± 2.1)U/mL,(72.2±1.2)U/mL,the cell density of them were 2.515±0.031,2.421±0.044.Starch was shown to stimulateα-Amylase production by B.subtilis,whereas glucose,sucrose was shown to inhibit that.
