Study on Absorption Mechanism of DFM by Using Everted Gut Sac Method

Objective To study the absorption characteristics of DFM(a component isolated from Radix Morindae Officinalis) in different intestinal segments of rats.Methods Everted gut sac method was used.The everted gut sac of different intestinal segments of rats was immersed into the Krebs solution containing 1mg/mL DFM.The solution inside the sac was collected and then drug concentration in the sac was determined by validated high pressure liquid chromatography-evaporative light scattering detector(HPLC-ELSD) method.Results DFM can be absorbed at all intestinal segments.The drug absorption constant(Ka) was in a decreasing sequence in the following intestinal segments: ileum duodenum jejunum.Conclusion There is no difference for DFM absorption in different intestinal segments of rats.
