Molecular mechanisms of inhibition of MyD 88protein to HBV gene replication

Objective To investigate the molecular mechanisms of MyD88 protein inhibiting HBV gene replication.Methods The adenovirus containing the MyD88 sequence is used to infect HepG2.215 cells,and immunofluorescence employed to confirm the efficiency infection of Ad5-MyD88.Adenovirus-GFP as negative control,after infection 48h,the HBV RNA,HBsAg and HBeAg were detected respectively by ELISA,Real-time PCR,Northern blotting and Southern blotting.The HBV core particle DNA also was detected via real-time PCR.All the results from real-time PCR were confirmed through Northern blotting or Southern blotting.Results Compared with negative control,the level of HBV RNA,HBsAg and HBeAg,Core particle DNA fell.Conclusion MyD88 can inhibit the HBV replication in HepG2.215.
