Effect of Prothymosin alpha Antisense Oligodeoxynucleotide on Human Gastric Cancer in Vitro.

Objective:To study the effects of Prothymosin alpha(ProT)antisense oligodexynucleotide(ProTα-AS-ODN)on human gastric cancer cells and to deploy a possible theapeutic approach of ProTα-targeted in human gastric adenocarcinomas.Methods: Antisense phosphorothioate oligodeoxynucletides targeted against ProTα mRNA were transiently transfected into BGC-823 cells to down-express ProTα.The altered expression of ProTα mRNA and protein were examined by RT-PCR and immunocytochemistry(ICC). Then,MTT assay and Hoechst 33258 staining were used to detect cytotoxicity and apoptosis.Results:The down-expressed ProTαmay induce a complete inhibition of cell proliferation to apoptosis in a dose-dependent manner in BGC-823 cells.Conclusion:The efforts aiming at cutting down ProTαexpression may prove to be an attractive alternative therapy against gastric cancer.
