Effects of recombinant human bone morphogenetic protein-2 on the proliferation and adhension of skeletal muscle satellite cells

2003
Objective:To investigate the effects of recombinant human bone morphogenetic protein-2on the proliferation and adhension of skeletal muscle satellite cells.Method:Skeletal muscle satellite cells were harvested and cultured in vitro.The0ng/ml,50ng/ml,100ng/ml,500ng/ml,1000ng/ml rhBMP-2were used to induce skeletal muscle satellite cells for48hours.The attached cells were calculated with fluorescence method at1h after seeding.Cell proliferation was measured with MTT method.Result:The number of attached cells have increased after the cells were induced with rhBMP-2when the concentration below500ng/ml,and reached the highest number when500ng/ml BMP.While the number did not increase with the increase of BMP concentration.rhBMP-2promoted cell proliferation when the concentration was above of500ng/ml.Conclusion:rhBMP-2enhances the adhension and proliferation of skeletal muscle satellite cells.Author's address Nanfang Hospital,First Military Medical University,Guangzhou,510515
