Expression of p33~(ING1b) and its Relationship with the Expression of p53 in Hepatocellular Carcinoma

Objective To investigate the p33ING1b mRNA expression level and its relationship with p53 in hepatocellular carcinoma(HCC)and to evaluate its clinical significance.Methods Reverse transcriptase-polymerase chain reaction (RT-PCR) method was used to detect the p33ING1b mRNA level in tissues from liver cancer and the adjacent lesion.The expression of p53 protein was also evaluated with immunohistochemical method.Results The expression of p33ING1b mRNA in cancer tissues (0.410±0.175)was significantly lower than that in adjacent tissues of cancer(0.529±0.203) (P0.05).The expression of p33ING1b was significantly correlated with the pathological grades 、lymph node metastasis and capsular infiltration,but not with sex、age、tumor size、tumor thrombus or liver cirrhosis.The expression level of p33ING1b mRNA in tumors With positive p53 expression(0.492±0.196) was significantly higher than that with negative p53 expression(0.363±0.176)(P0.05).Conclusion Down –regulated expression of p33ING1b may play an important role in the tumorigenesis and development of PHC.p33ING1b might be an useful marker in evaluating tumor differentiation 、invasion and prognosis.Abnormal expression of p33ING1b gene may be one of the mechanisms of p53 dysfunction in HCC.
