Reasearch of the Interaction of CPSF Proteins in Vitro

Object:The study was basd on the interaction of the four kinds of CPSF proteins and the interation of CPSF 73k,100k,160k and PAP in pre-mRNA 3′-end processing.Methods:The experiment of GST-pull down was carried out with the reticulocyte lysate translated protins of CPSF 73k,100k,160k and GST-CPSF 30k and GST-PAP which were bound to glutathione Sepharose~(TM) 4B.Results and Conclusion:The result showed that the four kinds of CPSF proteins can interact each other and involue the pre-mRNA 3’-end processing in the form of tetramer.Futhermore CPSF 73k,100k and 160k can interact with PAP.They all participate in the pre-mRNA cleavage and polyadenylation.
