Expression and biological function of cell proliferation and migration of linc00152 in human osteosarcoma

Objective To explore the expression and biological function of cell proliferation and migration of linc00152 in human osteosarcoma. Methods Real-time PCR was used to detect the differential expression of linc00152 in cancer tissue, normal bone tissue and SaoS2, U2OS, HOS, MG63 osteosarcoma cell lines and normal bone hFOB1.19 cell line. The expression of linc00152 in osteosarcoma cell lines was downregulated by small interfering RNA (siRNA), and the proliferation and migration ability of osteosarcoma cells was evaluated after knocking down linc00152 expression. Results The linc00152 relative expression in SaoS2, U2OS, HOS and MG63 osteosarcoma cells were 7.23±2.11, 5.02±1.89, 4.21±1.51 and 3.75±1.52 which was significant higher than normal bone cells hFOB1.19 (1.21±0.22) with statistical difference (F=9.358, P<0.01); linc00152 relative expression in cancer tissue and normal tissue were 4.12±1.81 and 1.97±1.03 respectively with statistical difference (t=5.490, P<0.01); The small interference RNA (si_00152) and unrelated sequences (si_nc) were transfected into the human osteosarcoma SaoS2 cells. It was found that the proliferation ability, migration ability and trans membrane ability of SaoS2 cells decreased significantly after down regulated linc00152 expression in SaoS2 cells. Conclusion Linc00152 increased significantly in human osteosarcoma tissues and cell lines, and participated in the biological process of cell proliferation and migration.
