Contribution of ATP-sensitive K~+ channel to emulsified isoflurane's protective effect on myocardial cells

Aim To discuss whether the ATP-sensitive K+ channel participates in the protective effect of the emulsified isoflurane(EI) on myocardial cells.Method The in-vitro neonate rat cardiac myocytes were primarily cultured,the H/R injury models were established and randomly divided into 6 groups,namely,normal control group(N group),H/R group(H/R group),H/R+fat emulsion group(F group),H/R+glibenclamide group(G group),H/R+1.68 mmol·L-1 EI group(EI group),and H/R+1.68 mmol·L-1 EI +10 mmol·L-1+glibenclamide group(EIG group).Supernatant liquid of cell culture was taken from each group(n=6) to mensurate LDH and cTnI,and myocardial cell homogenate was used to mensurate SOD and MDA;the changes in the characteristics and forms of myocardial cell growth were observed with inverted microscope.Results Compared to N group,the LDH,MDA and cTnI of other groups all increased(P0.05) and SOD declined(P0.05).Compared to H/R group,SOD of F group declined(P0.05),and the differences of the LDH,MDA and cTnI had no statistic significance(P0.05);the differences of G group's LDH,MDA,SOD and cTnI had no statistic significance(P0.05);LDH,MDA and cTnI of EI and EIG group declined(P0.05) and SOD increased(P0.05).Compared to F group,the LDH,MDA and cTnI of EI and EIG group declined(P0.05) and SOD increased(P0.05).Compared to EI group,the LDH,MDA and cTnI of EIG group increased(P0.05) and SOD declined(P0.05).Conclusion The protective effect of emulsified isoflurane(EI) on the primarily cultured myocardial cells hypoxia/reoxyenation injury may be associated with the ATP-sensitive K+ channel activated by it.
