Establishment of in Vitro Regeneration System of Poplar Leaves

The tender leaves of Populus euramericana 108 as the explants were cultured in the MS basic medium supplemented with different concentrations of plant growth regulators to establish the in vitro regeneration system.The results showed that the optimal induction medium was MS+2,4-D 1.0 mg/L+6-BA 1.0 mg/L and the medium suitable to induce the adventitious buds was MS+NAA 1.0 mg/L+6-BA 0.5 mg/L.NAA and BA had the regulation effects on the induction of calli,that was to say,the increasing ratio not more than 4∶1 was beneficial to induce the adventitious buds;while the relationships of BA with 2,4-D were more complex,the induction effect was the best with 2,4-D and 6-BA of 1.0 mg / L,respectively.
