Preparation and identification of monoclonal antibodies against avian encephalomyelitis virus

The Balb/c mouse was immunized with antigen of purified avian encephalomyelitis virus strain Van Roekel which was adapt for cell culturing. A hybridoma cell line, which can secret monoclonal antibodies against avian encephalomyelitis(AEV), was obtained. The characterization of mondonal antibodies was performed by chronosomal counting, specificity test, stability test and SDS-PAGE. The results showed that the chromosomal number was between 90 and 110, the subclass was IgG 2a ,the titer of supernatant was 1:1×10 4 , the titer of ascitic fluid was 1:1×10 6 ,the moleclular weight was 142KDa, and the secreting of monoclonal antobodies was stable, and no cross reaction was observed between the antibodies and the other avain viruses(NDV, EDSV,PMV 2 ,IBV,AIV) .
