Application of H_(22) cells stored in frozen to mouse model of BCG tumor-suppressing test

Objective To investigate the feasibility of application of liver cancer H22 cells stored in frozen to mouse model of BCG tumor-suppressing test.Methods H22 cells stored in frozen were prepared,of which those 10 and 83 d after storage were resuscitated and calculated for survival rate.The newly resuscitated H22 cells at high(7.5 × 106 cells / ml),moderate(5.0 × 106 cells / ml)and low(2.5 × 106 cells / ml)were mixed with 1 mg / ml therapeutic BCG respectively,with which BALB / c mice were inoculated subcutaneously,0.1 ml for each,using those with H22 cells at the corresponding dosages as control.The tumors of mice 30 d after inoculation were weighed,based on which the tumor-suppressing rate in test groups were calculated.The test was repeated.Results The survival rates of resuscitated H22 cells 10 and 83 d after storage in frozen were 85.45% and 82.25% respectively.The tumor formation rates in control groups were 100%.In the same test,the tumor weights were significantly lower in test groups than in the corresponding control groups(P 0.05),while showed no significant difference in various test groups(P 0.05).However,in different tests,the tumor weights of mice inoculated with H22 cells at the same dosages were significantly differen(t P 0.05).All the tumor-suppressing rates in various test groups were more than 60%.Conclusion The H22 cells stored in frozen met the requirements for tumor-suppressing test,while the stability of test needed to be improved.
