SIMULTANEOUS ESTIMATION OF CEFIXIME AND LINEZOLID IN BULK AND PHARMACEUTICAL FORMULATION BY HPTLC

Simultaneous estimation of Cefixime and Linezolid in bulk and pharmaceutical formulation by High performance thin layer chromatography (HPTLC) was successfully developed and validated. The chromatographic separation was carried out on pre-coated silica gel aluminium plate 60 F254 (20×20) with 250µm thickness (Merck, Darmstadt) using mobile phase comprised of methanol: ethyl acetate: ammonia (6.0: 4.0: 0.2 (v/v/v). The RF values for cefixime and linezolid were found to be 0.36 and 0.81 respectively. The linearity of the developed method was found to be within the concentration range of 150-1050 ng/spot for cefixime and 150-1050 ng/spot for linezolid. The LOD and LOQ were found to be 467.22, 1415.82 ng/spot for cefixime and 51.92, 157.33 ng/spot for Linezolid respectively. Validation parameters such as accuracy, precision, specificity, robustness and ruggedness were also established. The developed method was found to be simple, precise, specific and accurate.
