Optimization of Culture Mediums for Stock Spawn and Liquid Strain of Armillaria mellea Symbiotic with Gastrodia elata

A new medium(glucose 25%,yeast extract 2%,ethanol 0.5%,K2PO4 0.03%,MgSO4 0.015%,VB1 0.001% and agar powder 1.5%) for stock spawn of Armillaria mellea was obtained by orthogonal test.By using this medium,the growth of mycelia was superior to that of the widely used PDA solid medium both in biomass and traits.Another submerged culture medium(potato 8%,sucrose 0.8%,Castarea mollissima twigs 3%,chipped Corncob 3%) was also invented by adding Castarea mollissima twigs and corncob extracts into PDA medium.Compared to liquid enriched PDA medium,A.mellea in this medium can produce bigger,stronger mycelia and up to 2.5 times in biomass,and save 60% of potato and sugar use.Suitable proportions adding of Castarea mollissima or corncob extract could promote the growth of A.mellea,but Castarea mollissim extract is superior to corncob extract.The effect of combined two extracts is better than added separately,and the optimal ratio is 1:1.
