Changes of MMP-2,MMP-9,TIMP-1 in lung tissue of type 2 diabetic rats

2010
Objective To investigate the changes of MMP-2,MMP-9,and TIMP-1 in lung tissue of diabetic rats, and to explore the roles of MMP-2,MMP-9,TIMP-1 in lung injury of diabetic rats. Methods The experimental models of type 2 diabetic rats were established by injecting streptozotocin (STZ) and feeding high fat and glucose food, then the changes of morphology in the lung of rats were observed by light microscope at 12 weeks and 24 weeks respectively, and the collagen accumulation of lung was observed by masson trichrome staining ,and the levels of MMP-2,MMP-9 and TIMP-1 in lung were detected by immunohistochemistry. Results The amount of collagen fibre was increased obviously in lung tissue of diabetic rats, the expression of MMP-2,MMP-9,TIMP-1 and the ratio of MMP-2/TIMP-1,MMP-9/TIMP-1 was also increased in lung tissue of diabetic rats. With the progression of pathological changes, the changes became much more obvious. Conclusion There is pulmonary fibrosis in diabetic rats. The changes of MMP-2,MMP-9,TIMP-1 and the ratio of MMP-2/TIMP-1,MMP-9/TIMP-1 in lung tissue of dibtetic rats may be related to lung tissue pathological changes ,which may be one of the mechanisms for diabetic lung fibrosis.
