Effect of CD40-CD40 ligand interaction on the proliferation and migration in cultured cells of rat aortic smooth muscle

Objective:To investigate the effect of CD40-CD40 ligand interaction on the proliferation and migration in cultured aortic vessel smooth muscle cells(VSMC)of rat.Method:The primary passage smooth muscle cells of rats were cultured by tissue mince-sticking method and the 3rd and 4th passage cells were incubated with indicated concentrations of CD40 ligand(CD40L)for various periods.The incorporation of 3H-TdR and 3H-Leu analysis was used to evaluate the cell proliferation.Cell migration was obtained by monolayer cell scrape and observed by reverse microscope.Result:The incorporation of 3H-TdR,3H-Leu and the migration of smooth muscle cells were significantly increased by CD40L with a dose-and time-dependent manner.Anti-CD40 monoclonal antibody significantly suppressed CD40L-induced incorporation of 3H-TdR,3H-Leu and the migration of VSMC.Conclusion:The interaction of CD40-CD40L could promote the proliferation and migration of cultured aortic VSMC of rat.
