Drug concentrations in the pelvic retroperitoneal space exudate after the combined administration of cephems and astromicin

Abstract Cephems and aminoglycosides are frequently used incombination to enhance and widen the antibacterial spectrum. In this study, 200 mg of astromicin (ASTM) in combination with either 2 g of cefotaxime (CTX), 1 g of ceftizoxime (CZX), or 1 g of cefbuperazone (CBPZ) were administered to patients with uterine cervical cancer undergoing radical hysterectomy and pelvic lymphadenectomy. Concentrations of the administered drugs were measured in the exudate of pelvic retroperitoneal space and serum. Analysis of these concentrations using a twocompartment model showed that the pharmacokinetic parameters of ASTM, CTX, CZX, and CBPZ after combined administration are similar to those determined after administration of each drug individually. In the exudate of the pelvic retroperitoneal space, ASTM was detected at concentrations of 4.9 μg/mL or more, while CTX, CZX, and CBPZ were found at 11.1 μg/mL or higher concentrations. These results show that the combination of these drugs is useful for the treatment of infections in the field of obstetrics and gynecology.
