Study on preparation and in vitrotransdermal properties of Puerarin Microemulsion

Objective To prepare the Puerarin Microemulsion,and to study its quality evaluation and transdermal diffusion characteristics in vitro.Methods With oleic acid polyglycolic glycerol ester as an oil phase,polyethylene glycol-12-hydroxy stearic acid ester as a surfactant,and a glycol monoethyl ether as a co-surfactant,the aqueous phase was gradually added into the mixture containing drug to produce Puerarin Microemulsion.The morphology,particle size and distribution,stability and so on were studied.The transdermal characteristics of puerarin in Microemulsion and Ointment were compared by a modified Franz diffusion cell.Results With good stability,the appearance of the microemulsion was transparent.Its particle size was 30.1-56.3 nm,pH was about 5.6 and viscosity was 1.2 Pa·s.The steady-state skin flux of puerarin microemulsion and ointment were 2.28,and 0.75 μg·cm-2·h-1,respectively.Conclusion Puerarin Microemulsion penetration rate is about 3 times of the ointment.The microemulsion has a good penetration enhancing effect.
