An effectiveprocedurefor the synthesisof oligonucleotides by the phosphotriester methodhas been developed.The procedure is basedon the use of phosphateprotectinggroupsenabling 0-nucleophilicintramolecularcatalysisin the reactionof internucleotidebond formationunder the actionof arylsulfonylchloridesand theirderivatives.Using this new procedure,the time neededto performone elongationstepon polymersupportis 7-8 min. The effectivenessof the methodologyhas been demonstrated in the synthesesof many oligodeoxyribonucleotides of different lengthwith high yields.
