UJI SUHU SIMPAN DAN KEMASAN PUPUK HAYATI Rhizobium sp. CAIR

2018
The use of bio-fertilizers containing microbes often can not be used effectively. This is due to inadequate storage. Usually biological fertilizer is stored in a warehouse that is not equipped with a refrigerator, even when the temperature in the warehouse is high enough to make microbial viability on biological fertilizers to decline. The use of plastic as a packaging material has advantages over other packing materials because of its light, transparent, strong, thermoplastic and selective properties in its permeability to water vapor, O2, CO2. The nature of plastic permeability to water vapor and air causes plastics to play a role in modifying the container space during storage. Plastic is also a type of packaging that can attract consumer tastes. Most of the packaging of liquid fertilizer contained in the farm shop uses only one kind of bottle packing, so it is necessary to conduct research on several kinds of plastic bottle packaging that can be used for packaging of bio-liquid fertilizer. One alternative technology that can be used is the use of bio-fertilizers containing microbes. Thus, this study aims to test the storage temperature and packaging of Rhizobium sp. in the appropriate and effective liquid bacto pepton media prior to shipment. This research will be conducted at the Biotechnology Laboratory, University of Muhammadiyah Malang, East Java for 3 months (September-November 2017) using Split Plot design arranged by random group. The main plot is the different temperature of Rhizobium sp. liquid, S1 = 4 oC (in refrigerator), S2 = 25 oC (representing Kepanjen area) and S3 = 32 oC (representing Lamongan area) and sub plot is Rhizobium sp. liquid, K1 = glass bottle, K2 = clear plastic bottle (HDPE plastic type), K3 = blue plastic bottle (PET plastic type), K4 = brown plastic bottle (PET plastic type), and K5 = white plastic bottle (HDPE plastic type). Each treatment was repeated 3 times, so there were 45 experimental units. Each experimental unit consists of 5 samples. The result of observation is there are neither interaction and the real effect between the storage temperature and the packaging of Rhizobium sp. liquid. The yield tends to be highest at 25 oC storage temperature of 7,64x109 cfu / ml and plastic brown bottle packaging of 8,47x109 cfu / ml on the parameters of bacterial colonies. Then for color parameters of liquid biological fertilizer, bacterial colony form and bacterial velocity in forming colony, the treatment that given gives no significant difference.
