Bacteriostatic Activity of Bacteriocin from Streptococcus sanguis

Bacteriocin from Streptococcus sanguis was extracted and obtained by isolation, concentration, and purification with HA (hydroxyapatite) column chromatography, Sephadex G-150 gel chromatography, and ultra-filtration with hollow fiber column to study its bacteriostatic activity against suspicious pathogen of periodontosis using well-plate technique and Fusobacterium nucleatum as an indicator bacterium. The results showed that the purified bacteriocin has four mutual separated components, and protein of the II peak possesses bacteriostatic activity. Final production rate of bacteriocin after purification with freeze-dry was 0.082%, and solution of bacteriocin at 1 mg/mL formed an inhibitory ring with 17 mm in diameter, and its minimal inhibitory concentration was 62.5 μg/mL. Therefore, bacteriocin had a strong inhibitory function against suspicious pathogens of periodontosis.
