Kabak sarı mozaik virüsü izolatlarının kodladığı genlerin diziliminin belirlenmesi ve moleküler karakterizasyonu

Bu calisma, kabakgil alanlarinda problem olan kabak sari mozaik virusu (Zucchini yelllow mosaic virus–ZYMV) izolatlarinin kodladigi genlerin dizilimlerinin belirlenmesi amaci ile yapilmistir. Bu dogrultuda Ankara, Antalya, Burdur illerinden alinan enfekteli kabakgil bitkileri Zucchini yellow mosaic virus (ZYMV), Cucumber mosaic virus (CMV), Watermelon mosaic virus (WMV), Cucumber green mottle mosaic virus (CGMMV), Papaya ringspot virus-W (PRSV-W) ve Squash mosaic virus (SqMV) antiserumlari ile DAS-ELISA testine tabi tutulmustur. Test sonuclarina gore 233 ornegin 86’sinin (%36,9) ZYMV, 116’sinin (%49,7) WMV, 36’sinin (15,4) CMV, 22’sinin (%9) PRSV, 6’ sinin (%2,5) SqMV, 20’sinin (8,5) CGMMV ile enfekteli oldugu bulunmustur. DAS-ELISA testleri sonunda Ankara ve Antalya illerinde en yaygin virusler WMV ve ZYMV olarak belirlenmistir. Calismada ilk defa sakiz kabagi ve bal kabagi bitkilerinde CGMMV etmeninin varligi serolojik olarak tespit edilmistir. Test sonunda ZYMV ile enfekteli bitkiler secilmis ve Konya, Karaman ve Aksaray illerinden toplanan farkli kabakgillerden izole edilen ZYMV izolatlari ile birlikte molekuler olarak, P1 protein, yardimci bilesen protein (HC-Pro), P3 protein, silindirik inclusion protein (CI), Nuklear inclusion protein b (NIb) ve ortu proteini (CP) gen bolgelerinin baz dizilimi belirlenmis ve MEGA 6 bilgisayar programi kullanilarak filogenetik ve protein analizi yapilmistir. Calismada 53 izolatin gen bolgelerinin sekans verileri NCBI veri bankasinda yer alan sekanslarla karsilastirilmis ve 50 tane ZYMV Turkiye izolati Dunya’da ve Avrupa’da en yaygin olan A1 grubunda yer alirken uc tane ZYMV Turkiye izolati Avrupa’da yeni ortaya cikan A4 grubunda yer almistir. Bu sonuclarla Turkiye’de ilk defa ZYMV izolatlarinin gen bolgelerine ait bilgiler elde edilmis ve rekombinant olan izolatlarin varligi rapor edilmistir.AbstractIn this study, was performed to determine the sequences of the genes encoded by isolates of Zucchini yellow mosaic virus causing problem in cucurbits. Collected samples suspected to be infected from Ankara, Antalya, Burdur were tested with DAS-ELISA test to Zucchini yellow mosaic virus (ZYMV), Cucumber mosaic virus(CMV), Watermelon mosaic virus (WMV), Cucumber green mottle mosaic virus (CGMMV), Papaya ringspot virus-W (PRSV-W) and Squash mosaic virus (SqMV). According to the test results, samples were found to be infected with 86 ZYMV (36,9%), with 116 WMV (49,7%), with 36 CMV (15,4%), with 22 PRSV (9%), with 6 SqMV (2,5%) and with 20 CGMMV (8,5%). DAS-ELISA tests based on the results of Ankara and the most common viruses in Antalya province has been identified as WMV and ZYMV. In this study, CGMMV were determined first time in squash and pumpkins plants. Selected plants with infected by ZYMV and ZYMV isolates from different cucurbit plants obtained from Konya, Karaman and Aksaray, were sequenced the P1 protein, helper components protein (HC-Pro), P3 protein, cylindrical inclusion (CI), nuclear inclusion (NIB) protein and coat protein (CP) were determined according to sequence of the gene. Fifty three protein isolate sequence data were compared with sequences located in the NCBI data base. Fifty isolates from ZYMV Turkey were grouped in common group A1 and three ZYMV isolate Turkey took place in the newly emerging group in Europe A4.
