Expression of BDNF gene after gene transfer of AxCA-BDNF to the sciatic nerve for peripheral nerve regeneration in rats

Objective To observe the expression of BDNF gene after microinjected adenovirus-mediated gene transfer of BDNF (AxCA-BDNF) to the sciatic nerve for peripheral nerve regeneration. Methods Based on silicone tube graft as a support to bridge adult rat sciatic nerve gaps,Wistar rats were divided into 3 group receiving microinjection of recombinant adenovirus vector of BDNF (AxCA-BDNF), BDNF and simple injection of virus buffer to the sciatic nerve respectively. By using the methods of in situ hybridization and immunocytochemistry,the BDNF gene expression was detected. Results It was certified the BDNF gene was delivered into the Schwann cells and expressed efficiently. The expression of the BDNF gene lasted for 1 month, within the month nerve regeneration was accomplished in the rats. Conclusion The exogenous BDNF gene and its expression proteins were up-taken to the spinal cord mo-toneurons through retrograde axonal transport.
